Decreased Purine Synthesis during Amino Acid Starvation
نویسنده
چکیده
Normal human lymphoblasts starved for each of several essential, but not nonessential, amino acids had decreased DNA and RNA synthesis but no change in free intracellular purine nucleotides. The rates of purine nucleotide synthesis via the de novo and salvage pathways were measured by incorporating [14C]formate and [“C]hypoxanthine labels, respectively, into lymphoblasts starved for an amino acid or treated with a protein synthesis inhibitor. After 3 h of starvation, purine synthesis via the de novo pathway decreased 90% and via the salvage pathway decreased 60%. Cycloheximide and puromycin each reduced de novo synthesis by 96% and salvage synthesis by 72%. The decrease in purine synthesis de novo after removal of the amino acid was of first order kinetics and was fully and rapidly reversed by reconstitution with the amino acid. The synthesis of a-N-formylglycinamide ribonucleotide declined 97% after amino acid starvation; the synthesis of purines from 5-aminoimidazole-4-carboxamide riboside decreased 41%. The synthesis of guanylates decreased more than the synthesis of adenylates during amino acid starvation.

The aim of present experiment was to measure endogenous urinary purine derivatives using feed restriction method, as well as to determine the effect of 4 levels feeding on microbial protein synthesis. The present experiments was conducted by four camels about 326 Kg body weight. The camels were fed by different level of Atriplex leucoclada (AL), including maintenance, 1.25, 1.5 times of mainte...
متن کاملStringent control of fatty acid synthesis in Escherichia coli. Possible regulation of acetyl coenzyme A carboxylase by ppGpp.
The effect of amino acid starvation on the rate of fatty acid synthesis was examined in stringent (CP 78, ref+) and relaxed (CP 79, rel-) isogenic strains of Escherichia coli (leu-, his-, arg-, fhr-, fhi-). Rates of incorporation of [UJ4C]glucose, [lJ4C]acetate, and 3H,0 into chloroform-methanol-soluble lipids ( >95% phospholipid) were instantly reduced 50 to 60 % by leucine starvation of strin...

Deoxyadenosine (dAdo) and deoxyguanosine (dGuo) decrease methionine synthesis from homocysteine in cultured lymphoblasts; because of the possible trapping of 5-methyltetrahydrofolate this could lead to decreased purine nucleotide synthesis. Since purine deoxynucleosides could also inhibit purine synthesis de novo at an early step not involving folate metabolism, we measured in azaserine-treated...
متن کاملRole of methionine in the synthesis of nucleoside Q in Escherichia coli transfer ribonucleic acid.
Previously, we reported that starvation of Rel Escherichia coli for methionine, but not leucine or histidine, results in chromatographically unique species of aspartyl-specific transfer ribonucleic acid (tRNAAsp) lacking the modified nucleoside Q. The present studies demonstrate that methionine starvation of Rel+ E. coli yields a qualitatively similar, but less pronounced, effect. Furthermore, ...
متن کاملThe effect of amino acid starvation on nucleoside uptake and RNA synthesis in Tetrahymena.
The uptake of nucleosides and the synthesis of RNA in Tetrahymena thermophila were examined following amino acid starvation. Omission of leucine, phenylalanine, or arginine from the medium resulted in a rapid decrease in the incorporation of [3H]uridine into the acid-soluble pool and acid-insoluble material (RNA). Amino acid starvation inhibited the uptake of all ribo- and deoxyribonucleosides ...
